ANA773 — a novel Toll-Like Receptor-7 prodrug being developed as an oral therapy for cancer.

ANA773 is a novel prodrug of a proprietary TLR-7 agonist. Both the prodrug and its active substance were discovered, designed and synthesized by Anadys scientists. In preclinical studies, ANA773 has consistently demonstrated favorable biologic activity, including pharmacokinetics and induction of immunological response (pharmacodynamics).
In April 2007, Anadys presented data from an in vitro study showing that the active metabolite of ANA773 promotes natural killer (NK) cell-mediated anti-tumor response by inducing cytokine secretion, cytolysis of tumor cells and enhanced antibody-dependent cellular cytotoxicity (ADCC). In October 2007, Anadys presented preclinical data on ANA773 showing the impact that varying the dosing schedule has on the pharmacodynamic response to ANA773. Specifically, the report described that immunostimulatory responses to ANA773 in vivo can be modulated by schedule of administration.
The FDA has accepted our IND application to study ANA773 in cancer patients and we have recently initiated clinical trial sites. In the first trial we intend to explore the pharmacodynamic response in cancer patients. In the third quarter of 2008 we intend to select the tumor types for Phase 2 evaluation and in the fourth quarter of 2008 we intend to determine the dose and schedule for Phase 2 studies.
